Laser ablation in tadpoles
The team noted that femtosecond lasers had not been tried on Xenopus embryos, and tested them to address old questions about signalling during tail regeneration1. They tuned shutter time and power, settling on 200 ms and an average of 205 mW as one defined insult2. The paper's title states the finding: long-distance signals are required for tail morphogenesis.
Light-gated channels and tumours
Adams was a co-author of the 2016 study that expressed light-gated ion translocators alongside the KRAS oncogene in Xenopus. Light was applied to alter cell voltage. When the channel was stimulated inside developed induced tumour-like structures, 31% more embryos had normalized their tumours than among unstimulated injected controls3. Later reviewers cite Adams et al. (2013) for optogenetic hyperpolarization of tail stumps, which was reported to induce regeneration of a complete tail with a functioning spinal cord5.
Craniofacial work and citation
Adams, Levin and the artist Nataly Zahn published new Xenopus drawings in 2017. They were drawn from life, from several specimens, to average out variation. The authors contrasted this with Prijs, who worked from Faber's sketches4. A later review cites Adams et al. (2016) for craniofacial defects after disrupting the Kir2.1 homolog, which optogenetic changes in membrane voltage reproduced6. Funk cites Adams and Levin (2013) for distinct voltage domains within single cells7.
We decided to test the utility of fs lasers in Xenopus in the context of answering long standing questions about signaling during tadpole-tail regeneration.
